A Structured Illumination Method for Microscope Stage Tracking

This paper explores a new application of structured illumination to determine the position of a microscope stage. Measuring stage position accurately is important for any experiment that spans multiple fields of view. The structured illumination pattern is provided by a design printed on the microscope slide or cover slip that modulates the image of the specimen. A novel application of frequency analysis enables finding the orientation of the pattern and tracking the pattern independently from the specimen. We present results from synthetic and actual microscopy experiments and make an argument for a multiple camera system. The solution provides stage tracking in the presence of fixed and moving specimens, enabling measurements, tracking, and image alignment across multiple fields of view.
